Brief Introduction of L-Proline

L-Proline, often abbreviated as Pro or P, is a cyclic amino acid. Upon incorporation into peptide chains, it can undergo hydroxylation to form 4-hydroxyproline, a key component of animal collagen.

Hydroxyproline is also found in various plant proteins, particularly those involved in cell wall formation. Plants tend to accumulate significant amounts of L-proline under stress conditions such as drought, high temperatures, low temperatures, and salinity.

This amino acid is widely used in clinical applications, biomaterials, and various industries.

L-Proline exists in three forms: DL-proline, L-proline, and D-proline. L-proline is the naturally occurring form and crystallizes in columnar shapes at room temperature.

Plant Cold Resistance:
L-Proline is a component of plant proteins and is commonly found in free form within plant tissues. Under stress conditions such as drought or salinity, plants accumulate proline, which serves multiple functions:

    • Osmotic Regulation: Acts as an osmotic regulator in the cytoplasm.
    • Stabilization: Stabilizes the structure of biological macromolecules and reduces cell acidity.
    • Toxicity Relief: Mitigates ammonia toxicity and regulates cellular redox processes, acting as an energy reservoir.

Increased L-proline content in plants during adverse conditions (drought, salinity, heat, cold) reflects their stress resistance. Varieties with strong drought resistance typically accumulate more proline, making its measurement a useful physiological indicator for drought resistance breeding.

Additionally, proline’s hydrophilicity helps stabilize protoplast colloids and metabolic processes, reducing freezing points and preventing cell dehydration. Therefore, elevated proline levels in low temperatures enhance cold resistance and serve as a physiological indicator for cold resistance breeding.

Intrabiological Effects:
In living organisms, proline functions as an ideal osmoregulatory substance, a protective agent for membranes and enzymes, and a free radical scavenger, positively affecting plant health.
